Objectives:

The use of FCM in plants is influenced by endogenous fluorescence inhibitors and may cause an inaccurate estimation of genome size; thus, falsifying the relationship between genome size and phenotypic traits/ecological performance. Therefore, the optimized buffer that is tailor made in our analysis is used to minimise the errors. Use of fluorescent dye with flow cytometry enables relatively fast method which works on wide variety of materials and provides information on large number of nuclei. Relative fluorescence is used to calculate the genome size of the unknown plants from the family.

With the rapid expansion of whole genome sequencing and other genome related studies, there is an increase in the demand of user friendly and robust method of estimating eukaryotic genome sizes. Flow cytometry (FCM) is a commonly used method for estimating genome size in many organisms. It has become increasingly clear that variation in DNA amount has significant consequences not only at cellular, tissue or organism level but also at the ecological and evolutionary levels. It influences how, when and where a plant may grow and its chances of survival in the changing world.

What should be the idle choice of standards?
We use a set of different reference standards and standardization procedures (internal, pseudo-internal and external). Approach is to find the best reference standards and minimise the effect of inhibitors that is not limited to the sample type. Our goal is in addition to co-operations in the field of R&D to offer service on commercial base, e.g. for individual/ companies Institutions on this particular aspect.
Our methods established the use of Pongamia as one of the possible reference standard to estimate genome size of other potential biofuel crops.
