Isolation and Microscopic Visualization of T cells and B cells
Procedure
1. PBS is added to chicken blood sample
2. Blood-PBS sample is added to tubes containing histopaque
3. The tubes are centrifuged
4. Middle buffy coat is carefully pipetted out
5. PBS is added, and tubes are centrifuged
6. Supernatant is discarded and bottom layer containing cells is collected
7. Cells are placed on a clean and dry microscopic glass slide
8. Dye is added and incubated for 10 minutes
9. The slide is rinsed thoroughly with PBS
10. Visualize the cells under a microscope
